چکیده

Background and Aim: Sperm maturation occurs in the epididymis through interactions with existing molecules inside lumen. However, mechanism of molecular transfer is currently unclear. This study was aimed to determine necessity epididymal epithelial cells (EECs) process sperm terms kinetics tyrosine phosphorylation. Materials Methods: A true experimental research design used this study. The medium tested a primary culture mice caput (cells medium), conditioned (CM) (supernatant EECs), secretome (CM filtered at 0.22 µm). cocultured EEC culture, CM, for 1, 2, 3, or 4 h. original as control. kinetic analysis performed after indicated times using computer-assisted analysis, phosphorylation detected Western blot technique. Results: EECs successfully generated. results showed increased motility progressive movement 3 h incubation (p < 0.05). There significant decrease average path velocity (VAP) values 0.05), but there no change groups. culture-CM groups progressivity VAP percentage compared control In motility, CM were significantly different from medium, group not. Conclusion: cultured secretome, incubation, suggesting that can be replace EECs, especially when analyzing secreted by epithelium during maturation. highlight potential therapy mediums abnormalities. Keywords: kinetics,

Proteins from human oviductal tissue-conditioned medium modulate sperm capacitation.

BACKGROUND Spermatozoa acquire the ability to fertilize an oocyte when they become capacitated. Capacitation takes place when sperm pass through the female reproductive tract, interacting with female fluids. Both tyrosine phosphorylation of sperm proteins and the ability to respond to acrosome reaction (AR) inducers have been associated with sperm capacitation. Recent data indicate that conditi...

Characterization of epithelial cell culture from human hydrosalpinges and effects of its conditioned medium on embryo development and sperm motility.

BACKGROUND Recent studies have reported the negative impact of hydrosalpinx on IVF outcome. Toxic effects of hydrosalpinx fluid (HF) have been the main reason for the recommendation of functional surgery, salpingectomy, prior to IVF. The present study characterized hydrosalpinx epithelial cell culture and examined the effects of its conditioned medium (CM) on sperm motility, acrosome reaction a...

Sperm treatment affects capacitation parameters and penetration ability of ejaculated and epididymal boar spermatozoa.

This work was designed to study how this ability is affected by different sperm treatments routinely used for in vitro fertilization (IVF) assay. In this study, boar sperm samples from epididymal or ejaculated origin were processed by three different methods: left unwashed (NW group), washed in Dulbecco's phosphate-buffered saline supplemented with 0.1% BSA (BSA group), and washed on a Percoll(...

Study of TALP and TRIS citrate medium on caprine sperm capacitation and subsequent in vitro embryo production

The aim of the present investigation was to compare Tyrode’s albumin lactate pyruvate (TALP) medium and TRIS citrate medium for capacitation of caprine sperm. In experiment 1 capacitation was assessed by chlortetracycline assay and in experiment 2 with in vitro fertilization and embryo development. In experiment 2, cumulus oocyte complexes (COCs) recovered by slicing the caprine ovaries were ma...

Evaluation of cytotoxic effects of condition medium from amniotic epithelial cells on cancer cell lines HeLa and MDA-MB-231

Introduction: Amniotic membrane, the innermost layer of extra-embryonic tissue, contains mesenchymal and epithelial stem cells. The amniotic mesenchymal cells have the capability of inhibition of growth of cancer cells. In this research, the effects of amniotic epithelial cells on the viability of cancer cells and the role of apoptosis in this procedure were evaluated. Methods: Amniotic mem...
